After mitogenic growth factors interact with their respective membrane-associated receptors, phospholipase C(PLC) is activated, which lead to the hydrolysis of phosphatidylinosital 4,5-diphosphate to yield inositol 1,4,5-triphosphate and
diacylglycerol,
the physiologic activator of protein kinase C. Protein kinase C is believed to play a crucial role in tumor promotion Thus, PLC plays a central role in transmembrane signal transduction pathway. However, the clinical significance of its
expression
in
colorectal carcinoma is not fully established, We examined the relative content of PLC-¥ã1 and EGFR in human colon carcinoma by immunohistochemical staining of paraffin embedded sections. Among 26 cases, 19 cases(73.1%) demonstrated a significant
increase of PLC-¥ã1 content I comparison to the adjacent nontumorous tissue. The expression of PLC-¥ã1 did not correlate with Dukes stage and recurrence. The tumors that showed an increase in PLC-¥ã1 expression were exclussively EGFR positive
tumor.
These results suggest that PLC-¥ã1 is closely related to the EGFR and that PLC-¥ã1 way play an important role in colon carcinogenesis.
